A selective agonist of estrogen-related receptor gamma (ERRgamma), an orphan nuclear receptor involved in oxidative metabolism. Preclinical studies have investigated its potential as an exercise mimetic.
SLU-PP-332 is a synthetic small molecule that selectively activates estrogen-related receptor gamma (ERRgamma), an orphan nuclear receptor highly expressed in metabolically active tissues including skeletal muscle, heart, and brown adipose tissue. ERRgamma regulates the transcription of genes involved in oxidative phosphorylation, fatty acid oxidation, and mitochondrial biogenesis. By activating ERRgamma, SLU-PP-332 has been shown in preclinical models to shift skeletal muscle fiber composition toward oxidative type I fibers, increase mitochondrial content, enhance exercise endurance, and modulate metabolic parameters — effects that partially mimic the adaptations produced by aerobic exercise training.
Billon C, et al. Synthetic ERR-alpha/beta/gamma agonist induces an ERR-alpha-dependent acute aerobic exercise response and enhances exercise capacity. ACS Chem Biol. 2023;18(4):756-771.
Key finding: SLU-PP-332 produced an ERR-alpha-dependent acute exercise-like transcriptional response in mouse skeletal muscle and increased running capacity.

Key finding: Foundational work establishing the ERR family as regulators of mitochondrial metabolism and PGC-1alpha signaling, providing the biological basis for ERRgamma-targeted exercise mimetics.
