An extensively studied GLP-1 receptor agonist with a long half-life enabling once-weekly dosing. Research has investigated its GLP-1 receptor-mediated effects on metabolic signaling pathways and incretin pharmacology.
Semaglutide is a modified GLP-1 analog with 94% homology to native GLP-1. A C-18 fatty diacid chain enables albumin binding, extending its half-life to approximately 7 days. It activates the GLP-1 receptor in the hypothalamus and brainstem to modulate satiety signaling and food intake behavior, modulates gastric emptying kinetics, and enhances glucose-dependent insulin secretion from pancreatic beta cells. The compound also modulates glucagon secretion during hyperglycemia. These combined mechanisms produce sustained effects on metabolic signaling pathways and glycemic regulation.
