A synthetic growth hormone-releasing hormone (GHRH) analog conjugated with a Drug Affinity Complex (DAC) that extends its half-life to approximately 6-8 days. Studies have investigated its ability to sustain elevated growth hormone and IGF-1 levels.
CJC-1295 with DAC is a 30-amino acid GHRH analog (tetrasubstituted GRF 1-29) covalently attached to a maleimidopropionic acid-based Drug Affinity Complex. Upon subcutaneous injection, the DAC moiety forms a covalent bond with circulating albumin via a reactive thiol, dramatically extending the peptide's plasma half-life from minutes to approximately 6-8 days. This allows sustained activation of the GHRH receptor on pituitary somatotrophs, producing prolonged elevation of growth hormone (GH) and insulin-like growth factor-1 (IGF-1) levels without the sharp pulsatile pattern seen with native GHRH.

Key finding: Research investigated the pharmacokinetic and pharmacodynamic profile of CJC-1295 DAC, observing dose-dependent modulation of GH and IGF-1 signaling pathways that persisted over an extended duration.

Key finding: Despite continuous GHRH receptor stimulation, pulsatile GH secretion patterns were maintained, suggesting that CJC-1295 DAC preserves physiological GH release dynamics.
